Induced Thyrotoxicosis Elicits Protective Effects in Developing Hearts

Maternal hyperthyroidism effects development in infants and can lead to conditions such as craniosynostosis, exophthalmos, cardiac hypertrophy. Models for studying maternal hyperthyroidism, or thyrotoxicosis, are limited. Our lab has established an avian model study the of induced thyrotoxicosis on embryonic development. Thyroid hormones (TH) cause inotropic chronotropic changes tissue, including increased output, blood volume, Therefore, we wanted investigate our model. Fertilized chicken eggs injected days (E) 11 15 with either saline (control) 25ng thyroxine (T4). Hearts were harvested day E19, halves each sample used histological staining Masson’s Trichrome qRT-PCR analysis. The main objective was observe systemic by morphological genetic heart following exposure measuring expression THRa (TR receptor), ATP2A2 (calcium ATPases), MYH7(Myosin heavy chain 7). We hypothesized that levels all 3 markers would be upregulated, since THRaregulates transcription these other due fluctuating TH. These linked ventricular hypertrophy when upregulated. However, results contradicted hypothesis. There downregulation genes, a significant ATP2A2. In combination data collected related literature, suspect there is protective effect occurring hearts exposure.
